How the peptide calculator works
Enter the vial content in milligrams, the volume of bacteriostatic water you add in millilitres, and the amount you want to measure in micrograms. The calculator returns the concentration in mg/ml and the volume to draw in millilitres and in insulin-syringe units (U-100, where 100 units = 1 ml). It is a laboratory reference tool for preparing research solutions; it is not dosing guidance for humans or animals.
Worked example
A 5 mg vial reconstituted with 2 ml of bacteriostatic water gives 2.5 mg/ml, which is 2,500 mcg/ml. To measure 250 mcg you would draw 0.1 ml, which is 10 units on a U-100 syringe. Doubling the water to 4 ml halves the concentration to 1.25 mg/ml, so the same 250 mcg becomes 0.2 ml or 20 units.
Quick reference: 5 mg vial
| Water added | Concentration | 100 mcg | 250 mcg | 500 mcg |
|---|---|---|---|---|
| 1 ml | 5 mg/ml | 2 units | 5 units | 10 units |
| 2 ml | 2.5 mg/ml | 4 units | 10 units | 20 units |
| 3 ml | 1.67 mg/ml | 6 units | 15 units | 30 units |
| 5 ml | 1 mg/ml | 10 units | 25 units | 50 units |
Reconstitution notes for the laboratory
Add the diluent slowly down the inside wall of the vial and swirl gently; do not shake, as agitation can denature peptides. Label the vial with the date, concentration and batch number. Store reconstituted solutions refrigerated at 2–8 °C and protect from light; unreconstituted lyophilised vials are stored at −20 °C. Bacteriostatic water contains 0.9% benzyl alcohol, which is why it is preferred over sterile water for multi-draw research vials.
Frequently asked questions
How many units are in 1 ml?
On a U-100 insulin syringe, 1 ml equals 100 units, so 0.1 ml equals 10 units.
How do I convert mg to mcg?
Multiply by 1,000. A 5 mg vial contains 5,000 mcg.
Does the amount of water change the total peptide in the vial?
No. It only changes the concentration. More water means a larger volume is drawn for the same microgram amount.
Which syringe does the calculator assume?
A U-100 insulin syringe. If you use a different syringe, use the millilitre result instead of the unit result.
